Cell sorter

The MoFlo Astrios EQs (Beckman Coulter) is a high performance cell sorter that allows you to simultaneously separate up to 6 populations. It is equipped with two spatially separated lasers and with temperature control (sample to be separated and the sorted populations). It allows to use several collection tube types and to select different degrees of purity for each single population of interest.

Laser:

  • Laser 488 nm
  • Laser 640 nm

Detectors:

  • physical parameters FSC (possibility of measurement on both lasers) / SSC
  • 9 fluorescence parameters divided as follows:
    Fluorescence parameters available
    Laser 488 nm Laser 640 nm
    513/26 671/30
    576/21 722/44
    620/29 795/70
    664/22
    710/45
    795/70

Other:

  • adapters for the collection of sorted cells: 5 ml facs tubes, multiwell plates, 1,5-2 ml tubes, 50 ml falcon tubes, slides
  • temperature control both on the incoming sample and on the adapters in which the sorted cells are collected
  • availability of a biological hood in the cell sorter laboratory

  • Flow cytometry analysis on suspended cell colture samples
  • Measurement of the frequency/percentage of particular cell populations in a specimen labeled with specific antibodies
  • Separation and isolation of up to 6 immunophenotypically different populations simultaneously, favoring purity or recovery yield of target populations
  • single-cell sorting
  • Separation of cells of variable sizes, and of microorganisms (bacteria) and other small particles

Limited to Open lab staff.

  • Experimental design consultancy;
  • technical assistance;
  • the service is also open to direct collaborations on specific projects.

Access instructions for UniTo users:

  1. Read the Rule Book carefully.
  2. For each instrument for which access is requested:
    a) Fill in the specific “Access request” - UniTo users;
    b) Send to openlab.dsv@unito.it the Annex A present in the Rule Book, signed by the Operator and PI.
    Step a) and b) to be carried out simultaneously.
    Failure to receive the Annex A will make it impossible to book and use the required equipment.
  3. Only after receiving confirmation email from Open Lab staff (all sent data and documentations have to be verified):
    Book the instrument through the email openlab.dsv@unito.it.

Access instructions for NON-UniTo users:

  1. Read the Rule Book carefully.
  2. For each instrument for which access is requested:
    a) Fill in the specific "Access request" - NON-UniTo users;
    b) Send to openlab.dsv@unito.it the Annex B present in the Rule Book, signed by the Operator and PI, and document "Legge 136" (english version).
    Step a) and b) to be carried out simultaneously.
    Failure to receive the Annex B and the document "Legge 136" will make it impossible to book and use the required equipment.
  3. Only after receiving confirmation email from Open Lab staff (all sent data and documentations have to be verified):
    Book the instrument through the email openlab.dsv@unito.it.
