High Content Screening

The High Content Screening Platform (Nikon) is a system that combines automated immunofluorescence image acquisition and quantitative analysis to study biological mechanisms on fixed cells (slide) and viable cells (live imaging in multi-well plate).

 

  • Microscope Nikon Eclipse Ti2
  • Confocal Spinning Disk Crest X-lightV2: confocal system based on Nipkow's disc technology (instead of a pinhole as in laser scanning systems, there is a multi pinhole disc). Possibility of high-speed acquisition. Less impact on the sample due to LED lighting (better compatibility in live experiment). Equipped with the following excitation lengths: 395, 440, 470, 510, 550, 575, 640 nm. Fluorescence filters for DAPI, GFP, FITC and TRITC with bandpass filter. Field of view up to 25 mm.
  • Monochrome Camera: 95% quantum efficiency capable of capturing 63 fps at 12-bit with 2048 x 2048 pixel resolution.
  • Color camera: presence of CMOS sensor and 16.25 Megapixel resolution.
  • VCS Super-resolution module: resolutions equal to 120 nm on XY and 300 nm on Z axis
  • Perfect Focus System (PFS): device that controls and maintains automatically the focus

Objectives:

  • Fluorite 10x  (AN 0.3, WD 16.0mm)
  • Plan Apo 20x (AN 0.75, WD 1.0 mm)
  • Plan Apo 60x oil (AN 1.40, WD 0.13 mm)
  • Fluorite 40x ELWD (AN 0.6, WD 3.6-2.8mm, PH-2)
  • Fluorite 20x LWD (AN 0.7, WD 2.3-1.3mm, PH-2)

Other:

  • NIS element AR software with 2D / 3D tracking module, single particle tracking module, JOBS module for High Content screening.
  • Presence of incubator with temperature, humidity and CO2 control.
  • Adapters: multiwell plate, plates 35 mm ∅ (double-bay), slides (double-bay)

  • Ability to quickly analyze multiple parameters on cell culture plates and slide. It can be used for cell culture screening and assay development, in vitro enzymatic activity evaluation and toxicological and pharmacological assays.

  • the service is also open to direct collaborations on specific projects.

Access instructions for UniTo users:

  1. Read the Rule Book carefully.
  2. For each instrument for which access is requested:
    a) Fill in the specific “Access request” - UniTo users;
    b) Send to openlab.dsv@unito.it the Annex A present in the Rule Book, signed by the Operator and PI.
    Step a) and b) to be carried out simultaneously.
    Failure to receive the Annex A will make it impossible to book and use the required equipment.
  3. Only after receiving confirmation email from Open Lab staff (all sent data and documentations have to be verified):
    - For all DSV users and non-DSV users with a subscription: book the instrument from the Open Lab page through CampusNet platform with your SCU credentials.
    - For non-DSV user with a single use rate: book the instrument through the email openlab.dsv@unito.it.

 

Access instructions for NON-UniTo users:

  1. Read the Rule Book carefully.
  2. For each instrument for which access is requested:
    a) Fill in the specific "Access request" - NON-UniTo users;
    b) Send to openlab.dsv@unito.it the Annex B present in the Rule Book, signed by the Operator and PI, and document "Legge 136" (english version).
    Step a) and b) to be carried out simultaneously.
    Failure to receive the Annex B and the document "Legge 136" will make it impossible to book and use the required equipment.
  3. Only after receiving confirmation email from Open Lab staff (all sent data and documentations have to be verified):
    Book the instrument through the email openlab.dsv@unito.it.
