Confocal microscope
Leica SP8 is a next-generation laser scanning confocal microscope equipped with two detectors Hybrid (HyD) and a conventional detector for multi-channel scanning.
DM6 CS upright microscopes with HC PLAN s 10x / 25 eyepieces, fully motorized, equipped with DAPI, FITC and TRITC filters
Objectives:
- Air 10X (HC PL FLUOTAR, AN 0.3)
- Air 20X (HC PL FLUOTAR, AN 0.5)
- Water immersion 40X (HCX APO L, AN 0.80 W UVI)
- Oil immersion 63X (HC PL APO, AN 1.40 OIL CS2)
Confocal detection and scanning system:
- Detection system with 3 detectors (1 PMT e 2 HyD GaAsP)
- Conventional galvanometric scanning system (7 fps a 512x512 pixel),
- Resonance scanning system(28 fps a 512x512 pixel)
Laser:
- 405 nm Laser DMOD Flexible
- Argon Laser AOTF (458nm, 476nm, 488nm, 496nm, 514nm), 65 mW
- DPSS Laser AOTF 561nm, 20 mW
- HeNe Laser AOTF 633nm, 10 mW
Other:
- LAS X Lightning super resolution module that allow to reach a resolution of 120 nm in XY and 200 nm on the Z axis
- Navigator Software for mosaic constructions
- Incubator with temperature control, humidity CO2, with heating system for the lens
- High sensitivity multichannel scanning confocal imaging;
- Rapid sequential acquisition of fluorophores with minimal bleed-through or cross-talk;
- CO2 and temperature control for live imaging;
- 3D z-stacks;
- Mosaic reconstruction of large samples (Navigator function);
- Analysis of protein dynamics in live cells by FRAP / FRET.

Access instructions for UniTo users:
- Read the Rule Book carefully.
- For each instrument for which access is requested:
a) Fill in the specific “Access request” - UniTo users;
b) Send to openlab.dsv@unito.it the Annex A present in the Rule Book, signed by the Operator and PI.
Step a) and b) to be carried out simultaneously.
Failure to receive the Annex A will make it impossible to book and use the required equipment. - Only after receiving confirmation email from Open Lab staff (all sent data and documentations have to be verified):
- For all DSV users and non-DSV users with a subscription: book the instrument from the Open Lab page through CampusNet platform with your SCU credentials.
- For non-DSV user with a single use rate: book the instrument through the email openlab.dsv@unito.it.
Access instructions for NON-UniTo users:
- Read the Rule Book carefully.
- For each instrument for which access is requested:
a) Fill in the specific "Access request" - NON-UniTo users;
b) Send to openlab.dsv@unito.it the Annex B present in the Rule Book, signed by the Operator and PI, and document "Legge 136" (english version).
Step a) and b) to be carried out simultaneously.
Failure to receive the Annex B and the document "Legge 136" will make it impossible to book and use the required equipment. - Only after receiving confirmation email from Open Lab staff (all sent data and documentations have to be verified):
Book the instrument through the email openlab.dsv@unito.it.
